CD励起子法の新しい展開:単一官能基をもつ天然有機化合物への新規応用戦略

研究概要

1.有機化合物の絶対構造を非経験的に決定する方法として、「円二色性励起子カイラリティー法」が有用である。この方法の欠点は、発色団間の相互作用を使用するため、適用が複数の官能基を持つ系に限られ、monoalcoholなど単一の官能基を持つ系に適用できないことであった。我々は前年度、全く新しい概念と戦略に基づいて、単一の官能基を持つ系の絶対立体化学を「CD励起子カイラリティー法」で決定する研究を開始した。本年度は、以下の研究成果を得た。
di(1-naphthyl)acetic acidをキラルなモノアミンと縮合させてアミド体を合成した。このアミド体のCDを測定して、実際にどのような励起子分裂型Cotton効果が観測されるかを検証した。特に種々のアミノ酸メチルエステルの系では非常に強い励起子CDが得られ、立体配座の理論計算と組み合わせて絶対配置を明確に決定できる事を明らかにできた。
2.我々は最近2-メトキシ-2-(1-ナフチル)プロピオン酸(1:MαNP acid)が2級アルコールに対して驚異的な光学分割能を有し、1とアルコールから得られるジアステレオマーのNMR化学シフト値がナフチル基の磁気異方性効果により非常に大きくシフトすることを見い出した。そのシフト値から、アルコールの絶対配置が決定でき、新Mosher法としての応用が可能である。さらに、カルボン酸1は不斉中心が4級でありラセミ化が起こらないと言う利点を持つ。以上より、キラルカルボン酸1は光学分割と絶対立体化学の決定が同時に行える強力なキラルシフト試薬であり、MosherのMTPA、TrostのMPA、Riguera,KusumiらのMβNAを凌駕し、天然物の絶対配置決定に有用である。
