臓器移植における免疫関連因子のアンチセンスペプチドの応用

研究概要	同種、あるいは異種移植の拒絶反応、特に超急逝拒絶反応のメカニズムについて、分子生物学的な研究をまず行なった。特に異種間臓器移植では臓器の血管内皮細胞と、宿主の血液内の可溶性成分および細胞性成分の関連が主たる問題であることに着目した。すなわち、血管内皮細胞表面に存在する糖鎖抗原の分子解析と、その合成酵素であるガラクトシルトランスフエラーゼの遺伝子解析を行なった。そして抗原-抗体反応に基づく補体活性の誘導と、それに伴うサイトカインの活性、および細胞障害のメカニズム、特に細胞内のシグナルトランスダクションの分子生物学的解明が可能であった。超急性拒絶反応の制御に関しては、血管内皮細胞の表面に存在する糖鎖抗原の融解酵素であるガラクトシダーゼにつき、その遺伝子解析と、独自に開発したEndo-GalCの作成に成功した。さらにこの遺伝子を用いた遺伝子導入による超急性拒絶反応の制御効果につき、In vitroおよびIn vivoにおける実験を行なうことが出来た。同種移植における急性拒絶反応の制御の他の手段として、血液内の可溶性成分としてのペプチドを利用した免疫寛容の可能性を求めた基礎実験の成果が得られた。急性拒絶反応では宿主の細胞毒性リンパ球による、移植片のアポトーシスを介した細胞障害がその中枢としてのメカニズムであることを証し得た。特に、Fas抗原あるいはTNFによるレセプター型細胞死の誘導に関する分子について、Fasリガンドの一部の分子構造を利用したアンチセンスペプチドを合成し、それによる細胞障害の関連につき解明することが出来た。また、急性拒絶反応の制御としてこれらのアポトーシス関連分子の関わりの詳細を知ることが出来、臨床応用への可能性を示唆する研究結果が得られた。
