癌抑制遺伝子産物をターゲットにした新規の抗悪性腫瘍剤の開発

研究概要

ヒト乳癌培養細胞株
ER(+)の乳癌培養細胞株の代表であるMCF-7を用いて継代培養を行い、以下の実験を行った。
ヒト乳癌細胞株MCF-7中のNF2蛋白の定量
wetern blotting法によりヒト乳癌細胞中のNF2蛋白の同定をした。抗体は市販のNF2に対するポリクローナル抗体を使用し、NF2蛋白の存在を確認した。NF2蛋申は従来報告されている、分子量と同じ約78kDaであった。
組織化学染色による乳癌組織におけるNF2蛋白の発現確認
NF2のポリクローナル抗体を用いて、乳癌組織の免疫組織化学染色を施行した。その結果、25%の症例においてNF2強陽性であった。強陽性の症例は進行した病期の割合が多く、陰性の症例は病期1以下が多かった。
NF2のカルパインによる分解、カルペプチンによる分解阻害
以前に行った実験と同様にヒト乳癌細胞株MCF-7培養液中に種々の濃度のカルパイン阻害剤であるカルペプチンや、他のカルパイン阻害物質を添加し、細胞増殖を観察した。その細胞をwetern blotting法により、カルパインの活性化の程度、カルペプチンを始めとするカルパイン阻害剤によるカルパイン活性化の阻害は濃度依存性に抑制された。同様にNF2蛋白の分解阻害も認められた。
新規抗癌剤の効果予測をするための感受性試験CD-DST法の乳癌症例における検討
乳癌に感受性の高いCyclophosphamide+Epirubicin(CE)あるいはDocetaxel(DOC)を用いた、抗癌剤感受性試験を実際の乳癌臨床材料を利用して、Collagen Gel Droplet Embedded Culture-Drug Sensitivity Test(CD-DST)法により施行、新規抗癌剤の開発に備えた。その結果、正診率はCEとDOCでそれぞれ、85.7%と96.4%であり十分に利用可能と考えられた。
