研究概要

本研究は、細胞バリアの本体であるタイト結合の機能と構造の理解を通じ疾患におけるタイト結合の役割を明らかにし、治療や薬剤投与の新しい理論基盤を形成すること究極の目的とする。
本年度はBarmotinと他の関連分子のクローニングを中心に研究を勧めた。加えてタイト結合の生理機能および分子構造における各種タイト結合関連分子の機能について検討した。
(1)Barmotinの構造と機能の解析
Barmotinの1次アミノ酸構造の詳細な解析により本分子にはF actin結合ドメインとPDZドメインを有することが示唆され、これらのドメインの機能を検討するために組換えF actin結合ドメインとPDZドメインを作成し、結合機能を解析した。
(2)タイト結合のバリア機能とフェンス機能のアクチンによる制御機構
タイト結合のバリア機能とフェンス機能が、アクチンにより異なる制御機構を介して制御されている可能性を、新規アクチン脱重合剤ミカロライドBによって解析した。
(3)Occludinによるcortical actinの制御
occludinの発現により血管内皮細胞のcortical actinが制御していることを見出し、タイト結合の機能に重要や役割を果たしていることを見出した。
