野生霊長類の薬用食物の探索とその生理機能性成分の解明

研究概要	薬を持たない野生霊長類では食を通して疾病の治癒・軽減や予防をしているものと考えられる。本課題は、タンザニア・マハレ山塊国立公園の野生チンパンジーを対象に、(1)非栄養的摂取植物の生理機能や活性成分の究明と、(2)これら植物の採食意義を解析すること、を目的としている。この研究は、薬化学や食品機能学、さらには動物生態学に寄与するものである。平成11年度の現地調査にて、非栄養的採食植物約20種を採取した。平成12年度では、これら植物の生物・生理活性を総合的に評価し、特徴ある種に含まれる生理活性成分の研究に取り組んだ。野生動物の薬として最重要との観点から実施した抗寄生虫スクリーニング(抗住血吸虫活性)では約半数が有意な活性を示した。なかでもTrema orientalisのそれは強く、チンパンジーの本植物採食における特徴からも興味がもたれ、一活性物質の精製単離に成功した。巾広い生理活性が検索できる試験系、殺塩水エビスクリーング、ではSenna spectabilitisおよびAnchomanes dubisに顕著な活性を認め、現在これらの活性成分の研究を展開中である。上記植物類は機能性食品学的観点からも貴重であり、T.orientalisには抗ラジカル産生抑制活性が、またS.spectabilisには抗ラジカル産生と共に発がん抑制活性が認められた。抗ラジカルや発がん抑制の観点からはさらに有意な種を指摘できた。また、派生的研究として、民間伝承種(Anagalis arvensis)の住血吸虫中間宿主制御物質や、東南アジアの野生猿類の採食種の生理活性スクリーニングも行った。さらにまた、チンパンジーの食(味)と薬(生理機能)との関連を野外で解析する一アプローチとして、渋味の塩化第二鉄反応を利用した定量化試験法を試行し、現在その妥当性を解析中である。また、平成12年度には、現地ダルエスサラーム大学化学科よりDr.Nkunya教授を招聘し、将来へ向けた研究体制作りを行った。
