T細胞特異的な細胞周期制御機構の解析

研究概要

細胞増殖、特に細胞周期の研究は多くが繊維芽細胞で行われて先行している。細胞周期機構の基本はリンパ球にも適用されるが、細部については違いがみられる。我々は細胞周期の転写因子E2Fに着目して解析し、繊維芽細胞ではE2F1の過剰発現により細胞周期がS期まで進行するが、ヒトT細胞株ではE2F1の活性上昇も認められなかった。そこでT細胞の増殖因子であるインターロイキン2(IL-2)によるT細胞増殖のE2F活性化機構を詳細に調べた。IL-2依存性ヒトT細胞株Kit225の培養からIL-2を除き休止期に同調した。再び、IL-2を加え細胞周期を進行させるとS期、G2/M期の細胞が15%から64%に増加した。それに伴いE2F1の発現上昇が認められた。さらに、E2Fの活性化状態を調べると、IL-2刺激により休止型のE2F4/p130の複合体が減少し、遊離型のE2F4が増加していた。遊離型のE2Fは休止期の細胞ではみられない。遊離型E2F4は活性化E2FでDNA結合能を示した。また、休止型複合体にかわってS期複合体が増加した。S期複合体はp130、p107、E2F4、Cdk2サイクリンAを含んでいた。IL-2刺激はp130とともにpRb、p107のRbファミリー分子のリン酸化を引き起こし、その結果、E2F4からの解離が生じ活性型のE2F4が出現したと考えられる。現在IL-2によるCdkリン酸化酵素の活性化を検討している。
