T細胞特異的な細胞周期制御機構の解析

研究概要	細胞増殖、特に細胞周期の研究は主に繊維芽細胞で行われ多くの成果が蓄積している。細胞周期機構の基本はリンパ球にも適用されるが、細部については違いがみられる。E2F1の過剰発現により繊維芽細胞では細胞周期がS期まで進行するが、ヒトT細胞株では細胞周期の進行は誘導されずE2F1の活性上昇も認められないのが一例である。そこでT細胞の増殖因子であるインターロイキン2 (IL-2)によるT細胞の増殖を、昨年に引き続き細胞周期の観点から解析した。IL-2はE2Fを活性化して細胞周期をS期、G/M期に誘導し細胞増殖を促す。E2Fの活性化は、pRbファミリーがリン酸化されpRbファミリーがE2Fから遊離することによって生じ、そのリン酸化はcyclin-dependent kinases (CDKs)による。IL-2をT細胞に作用させると、CDK2、CDK4、CDK6の活性が誘導される。CDKsの活性化はCDK自身の遺伝子の発現がIL-2に誘導されることと、cyclin D2の転写発現誘導によるこれらの分子の発現量の増大が原因の一つであることが明らかとなった。またIL-2はCDK inhibitorsの内P19とP27を蛋白レベルで減少させることを見い出した。以上の観察より、IL-2はT細胞で、CDK inhibitor活性の低下とCDK/cyclin複合体の増加によりCDKの活性化を引き起こし細胞周期を進行させるものと考えられる。
