ヒトにも活性な昆虫成長因子GBPを受容するレセプターの蛋白質構造解析

研究概要

早川らは、寄生蜂が宿主に卵を産みつける際、宿主に誘導するペプチドであるGBPを発見した。GBPは高濃度で宿主幼虫の成長を抑制し、寄生蜂幼虫の成育環境を守るが、低濃度では逆に細胞成長因子他いくつかの機能を示すサイトカインでもあることが分かってきた。またGBPはヒトケラチノサイトに対してもEGFと同等の増殖活性を有していることが分かっているが、分子量がEGFの半分しかないGBPがEGFレセプターと結合し信号を細胞内に伝えることは驚くべきことである。本研究の目的は、GBPとレセプターの結合状態の立体構造解析を行い、その作用メカニズムを明らかにすることである。
GBPレセプターについては、全長配列の決定を行った。現在、発現系を構築している。
EGFレセプターの膜外ドメインについては、海外の共同研究者を得て発現精製のメドがたった事から、GBPとの相互作用解析を行なう。
宿主体液中には、GBPと結合する可溶性の蛋白質が発見されており、この蛋白質とGBPの結合もGBPレセプター系のモデルとして興味深いので構造解析を行う。現在大腸菌による発現系を構築して蛋白質を得ており、BiacoreによるGBPとの相互作用解析を行なっている。
リガンドの残基置換による活性変化からもレセプターとの相互作用を検討している。N末端およびC末端数残基をdeletionすることにより、細胞増殖促進活性と血球活性化活性が各々異なった影響を受けることから、両活性を発現するレセプターは別の分子であることが推定された(J. Biol. Chem.,24,31813-31818(2001))。またGBPのY11A変異体が2次構造をもたないにもかかわらず活性を有することから、TFE中ではGBPと同じ構造をもつ事を明らかにし、レセプター結合によるリガンド構造の誘導を示唆した(J. Biol. Chem.,in press)。
