研究概要

副腎皮質刺激ホルモン(ACTH)に焦点をしぼり、分泌顆粒の形成と細胞内輸送、特に分泌に関わる新規因子の同定と抑制を目的とし、ACTH恒常的過剰発現細胞株を新規に作製した。蛍光タンパク（GFPとその変異型をさす）を融合した野生型ACTH cDNA、splicing variantである大分子量ACTH cDNAをepisomal vectorを用いて、AtT20細胞株へ導入した。共焦点レーザー顕微鏡を用いて、ACTH分泌顆粒の細胞内局在を観察した。CRHや細胞外高カリウム刺激で、ACTHの分泌が生じることを観察し、細胞膜直下の分泌顆粒の動態を、全反射顕微鏡を用いて観察した。
