Outline of Annual Research Achievements

本研究では、SASPを阻害する天然物を見出し、慢性炎症を克服する戦略及び新規予防標的分子の発見を目的としている。ヒト線維芽細胞にブレオマイシンを加えることで細胞老化を誘導し、細胞外に分泌されるIL-6をELISA法によって測定するSASP評価系を構築した。SASP阻害天然物のスクリーニングを行った結果、延命草成分オリドニンにSASP因子の産生抑制能を確認し、その機序についてNF-κBやp38の関与が認められた。さらに、自動分注機を用いたスクリーニングの系を構築し、複数の新たなSASP阻害候補物質が得られた。これらの候補物質について、SASP阻害機序の一部を明らかにした。また、文献上SASP阻害が報告されているものの、その詳細な機序が不明な化合物Xに着目し、化合物Xと結合する複数のタンパク質の同定に成功した。siRNAを用いた検証により、結合タンパク質の一つにSASP阻害との関連が示唆された。
またSASPを起こした細胞は活性化NK細胞により積極的に攻撃・排除されることから、NK細胞を活性化することでSASPを阻害する物質を探索した。そのためにT-bet結合配列をつないだレポータープラスミドを安定発現する細胞株を樹立し、この細胞を用いたハイスループットスクリーニング系を用いてNK細胞を活性化する物質を探索した。その結果、天然物エキスと認可済み薬剤からT-betの転写活性化を誘導する候補天然薬物を同定し、さらに数種のリード化合物を取得した。天然物エキスにおいてはT-bet活性化に関わる成分解析を行い、候補化合物の同定を行った。また3種類のリード化合物について、NK細胞活性化機序の解析とin vivoにおける薬効の検証を行った。SASPを抑制するNF-κB阻害作用を有する天然物由来化合物を多数同定し、これらのin vivoにおける転移抑制作用について明らかにした。
