Research Abstract

生体分子の構造揺らぎを考慮した自由エネルギー計算手法の開発
溶液中の化学プロセスを理解する上で鍵のひとつとなるのが自由エネルギーの計算である。分子認識に関連する結合定数、生体分子の構造安定性の議論など、自由エネルギー計算の応用例は枚挙にいとまがない。既存の3D-RISM理論に基づく自由エネルギー計算は、基本的に分子の内部自由度を凍結するという前提の下で行なわれてきた。これに対し本研究では3D-RISM理論に基づきつつ、溶質分子(生体分子等)の揺らぎを考慮した自由エネルギー計算を目的とした。この目的に沿って、3D-RISM理論と分子動力学(MD)シミュレーションの結合手法[1]を用いた自由エネルギー計算法を提案した。
自由エネルギー計算はオーダーパラメタの選び方によって、様々な手法が考えられるが、ここでは(1)カップリングパラメタ、および(2)分子間距離、をオーダーパラメタとする二つの熱力学式積分法を提案した。
これらの手法は薬剤設計において重要となる「蛋白質の構造揺らぎを考慮した分子認識過程の解析」に威力を発揮することが期待される。
