Research Abstract

これまで申請者らは、Rab27aおよびそのエフェクターgranuphilinが、膵β細胞インスリン顆粒の細胞膜ドッキングと融合の制御に重要な役割を担うことを示してきた。しかしその後、全反射顕微鏡を用いた、生きた膵β細胞における顆粒の開口放出現象の観察により、分泌様式は必ずしも一様ではなく、これを裏打ちする複数の分子機序の存在が示唆された。そこで本研究では、調節性分泌経路で働く低分子量GTPase Rab27a/bとそのエフェクター分子exophilinsを中心とした、分泌小胞開口放出に関する分子基盤を解明する。この目的のために、(1)Rab27a、granuphilin等に結合する分子群のプロテオミクス解析による網羅的同定、(2)生細胞における、小胞膜局在分子の分泌小胞細胞内動態に及ぼす影響の解明、(3)機能解明が進んでいないRab27エフェクター、およびこれらと相互作用する分子に関する遺伝子欠損動物の作製・表現型解析、を行う。
