本研究では、共生体によって駆動される真核生物の進化の基本原理を解明するという本研究領域の全体構想の中で、オルガネラの創成と進化の理解に根ざしたオルガネラの人為的操作を通して有用生物を創出すること、すなわち人工マトリョーシカ創成への試みを実
施している。
平成27年度においては、洲崎は細胞内共生中のクロレラからPV膜を単離し、含まれるタンパク質を解析した。RNA-Seq法により得られたmRNAデータを基にPV膜に存在するタンパク質を網羅的に同定し、その特徴を解析した。さらに、PV膜の起源である食胞膜についても同様に網羅的プロテオーム解析を行い、PV膜を特徴付けるタンパク質成分を質量分析法により同定し、それらの機能解析を行った。その結果、クロレラが産生したマルトースをホストの細胞内へと輸送するための輸送体、ホストからクロレラへのアミノ酸の輸送体、PV膜内部を低pHに維持するためのV-type ATPaseを含む、多くのタンパク質が同定された。
橘は、オルガネラ移植後のレシピエント細胞を継時的に観察し、移植マイトソームの変化を解析した。またAtg8遺伝子の発現を抑制した細胞をレシピエントに用い、移植マイトソームがオートファジーによって分解される可能性について検討した。さらに、移植オルガネラに対するレシピエント細胞の許容度を評価するために、Entamoeba nuttalliへのマイトソーム移植を実施した。
