Outline of Annual Research Achievements

「分子ほぞ」という新しい化学結合の概念を実践し、数種類の歯車状両親媒性分子(GSA)を合成し、自己集合を調べた。その結果、分解温度が130 °Cと水の沸点をはるかに超える高い安定性を示す箱型六量体「ナノキューブ」を開発した.また、このナノキューブに存在する1 nmの空隙を疎水分子で充填すると、分解温度が150 °Cを超え、超好熱菌タンパク質の中で最も安定なパイロコッカスホリコシイCutA1の持つ148.5 °Cを超えた.続いて、分子のかみ合いを可視化する新手法(SAVPR: Surface Analysis with Varing Probe Radii)を開発した。これはコンピューター上で分子表面を求めるプローブ球の大きさを変化させ、接触面積の面間距離分布を求める手法である。SAVPRにより一連のナノキューブのかみ合いを調べた結果、ナノキューブの安定性は分子のかみ合いと良い対応関係があり、ナノキューブがvan der Waals力により大きく安定化されていることがわかった。
自己集合性錯体の形成過程を我々が開発したQASAP (Quantitative Analysis of Self-Assembly Process)により調べた。Pd12L24錯体では、PdnL2n (n = 6, 8, 9)の閉じた構造とNMRで観測できない中間種が生成し、これらが最終構造へ至った。柔軟性の異なる二座配位子からなるPd2L4かご構造では、柔軟性が高いと、一過的に二次元シートを形成し、これが分解しかご構造へ変化した。また、Pd4L8四面体型、Pd4L8 double-walled triangleの形成過程も解明し、数百nmの中間体を一過的に生成することがわかった。また、キラルセルフソーティングの機構を世界で初めて解明し、自己集合のどの段階でソーティングが起こるかを突き止めた。
