Research Abstract |
Breeding by cell fusion is excelent method, especialy in breeding program among species and/or genus. It's not neccesary to consider for commpatibility among them. In this research, it tried to transfer cytoplasms related male sterile in rice plant (Oryza sativa L.), and to use cybrids gotten in this process were used for analysis of expression mechanism of cytoplasmic male sterile. Cell fusion were done by assinmetric, and fusion combination was between fertile and sterile. 1. Cybrids showing cytoplasmic male sterile were examined their mitochondrial genome by mtDNA cosmid clone covering all sequence. Two dmains were closely related for seed fertility of cybrid. One domain is including atp6 gene, and other's is located rps1 gene. This is the new information for gene related cytoplasmic male strile. 2. For variable characters of wild rice having cybrids, survey of blast desease does not show higher resistance than O.sativa. 3. Some sterile cybrids were grown fertile and sterile tillers through stock storage, and for these differences between fertility, the analysis of mitochondrial genome for these differences were coresponding to above two domains.
