| Research Abstract |
(1)Williams症候群患者血液に、SV40を感染させ、Williams症候群の細胞株を確立した。確立した細胞株をfluorescent in situ hybridazationで検討した所、染色体の異常が認められた。(2)Williams症候群患者血液中の白血球、及びその細胞株から、RNAを抽出し、カルシトニンのプライマーを用いて、RT-PCR-ダイレクトシークエンスを試みたが、欠損部が存在する為か、全塩基配列の確認には至らなかった。(3)そこで、ダイレクトシークエンスが可能であった部位の内、もっとも下流域に相当する部位から、5′Rapid Amplification of cDNA Endsを行い、cDNAクローンを得る事に成功した。(4)このクローンから、ダイレクトシークエンスを行った所、遺伝子バンクに存在しない塩基配列であった。(5)但し、RACE法は、PCR反応を基本とするので(PCR反応に付き物のア-ティファクトが起こり得るので)、念の為、現在、このクローンをサブクローニングし、塩基配列の同定中である。(6)さらに、cDNAライブラリーを作成する事で、配列を確定する予定である。
