| Research Abstract |
EBウイルス増殖のためのパッケージング細胞開発を行った。EBV DNAは約172キロ塩基対(kbp)の長さである。EBV産生に際してEBV DNAがウィルス粒子内ヘパッケージングされるためには、ウイルスDNAのサイズは150-200kbpの範囲になければならないことが明らかとなっている。そこで、EBV産生細胞株であるAkata細胞のウイルスDNAへ約50kbpの長さの外来DNAを組み込んだ。得られた細胞では、ウイルス産生を誘導してもウイルスDNAが大きすぎるためパッケージングされず、感染性のウイルス粒子が産生されないことが期待される。このような細胞へEBVの複製起点配列とパッケージングシグナル、外来遺伝子を含むDNAを導入すると、コンカテマーを形成して、パッケージングに適当なサイズで切断されて、ウイルス粒子中に取り込まれることを確認した。
