| Research Abstract |
インフルエンザウィルスは、宿主のシアル酸を含む糖鎖を受容体として認識し特異的に結合するヘマグルチニンタンパク質と自らの受容体を破壊するシアリダーゼを併せもつ極めてユニークなウィルスである。このウィルスによる感染は、3量体として分布しているヘマグルチニンによる宿主のシアリル化糖鎖への結合によって引き起こされる。すなわち、この初期段階のヘマグルチニンと宿主細胞との結合を阻害する化合物の合成を行った。目的とした化合物は、上述の多価型受容体に効果的に結合することが期待できるデンドリマー構造の化合物群を選定した。初年度は、以下のことを中心に合成と評価を行った。
★糖鎖構造を担持するためのコアとなる有機ケイ素デンドリマーを世代と形状に着目して系統的に合成した。(投稿準備中)
★これまで我々が利用してきた液体アンモニア中でのワンポット結合反応が、種々の官能基を有する単糖由来の糖鎖誘導体と有機ケイ素デンドリマーとの結合反応についても展開可能であることを見出した。(Carbohydrate Res.誌に掲載済み)
★デンドリマーコアへの導入のための前駆体となるシアリルラクトース誘導体を化学合成した。(投稿準備中)
我々の合成戦略は、原料を厳密に選定することにより、これまで極めて合成例の少ない正確な分子量を有する多価型糖鎖リガンドの分子形態を系統的に整えていくユニークな手法であり、次年度以降の展開が期待できる。
