| Research Abstract |
survivin anti-senseとして機能するEPR-1 cDNAをmetallothionein promoterを持つPlasmid vector(pMTN2)にligateしたpMTN2-EPR-1 plasmid vectorをヒト大腸癌細胞HT29(survivin発現陽性)に導入して得たEPR-1高発現株HT29-AS33,-AS49がzinc投与によってアポトーシスが強く促進され、Cis-DDPならびに5-FUに対する抗癌剤感受性が著明に増強されることをin vitroにおいて確認した。ついで、将来の臨床展開を踏まえてEPR1を含むAdenovirus vector(Ad. CMV-EPR1)を作成して、HT29ヌードマウスxenograftsに対するin vivo治療効果を検証した。HT29 xenograftsが8mm径以上となった担癌マウスに対するAd. CMV-EPR1ならびにAd. CMV-LacZの一回投与は後者に比べて前者が1/4の腫瘍増殖抑制を来たし、その効果がHT29細胞のアポトーシス誘導によるものであることを明らかにした。加えて、CDDPあるいは5-FUとAd. CMV-EPR1投与の併用により治療効果が増強され、前者では25%、後者では16%の腫瘍消失を来たした(Eur J Cancer, in press)。また、研究代表者らはかねてからdThdPase(TP)cDNAのplasmid vectorを介したtransfectant cellsが5'-DFURや5-FUの感受性増強に関与することを明らかにした(Br J Cancer 75,1997)が,本研究においてはTPcDNA(1.5kb)をレトロウィルベクターpNV7のEcoRI siteに挿入し、マウス大腸癌細胞株MC38-TPならびにMC38-Neoを得た。Western blotやELISAにて前者のみの有意なTP発現が認められること、両細胞の増殖活性に差のないこと、5-FU,5'-DFUR, Capecitabineに対するIn vitro感受性が前者において著明に増強していることを確認の後に、それぞれのマウス皮下移植腫瘍に対するin vivo感受性を検討し、MC38-TPの5'-DFUR, Capecitabine感受性が著明に増強されていることが確認された(in submission)。こうした感受性関連遺伝子による治療研究は現行の癌化学療法の効果向上に向けて有用な知見を提供している。
