Research Abstract |
In order to develop a successful ex vivo gene therapy to enhance healing potential of pulp tissue and induce reparative dentin formation after injury such as caries, we developed a three-dimensional pellet culture system of pulp stem cells electrotransfected with growth/differentiation factor 11 (GDF11). The viability after electroporation was more than 85%, and the efficiency was around 65% calculated by flow cytometry. Real time RT-PCR analysis showed that the expressions of alkaline phosphatase, Dentin matrix protein 1 and Dentin sialoprotein, a differentiation marker of odontoblasts were increased in the pEGFP-Gdf11 transfected pellet compared with those in the pEGFP transfected pellet. The autogenous trasplantation of Gdf11 transfected pellet on the canine amputated pulp stimulated the reparative dentin formation during pulpal wound healing. Therefore, the ex vivo gene therapy using this pellet culture system has the potential for regeneration of exposed pulp with a large amount of dentin as an endodontic treatment
