Research Abstract

In this project, we tried to improve the tissue engineering technology for production of cultured skin using developmental and regenerative molecular mechanism. We developed a new technology to accelerate the formation of the cultured epithelium by using Sonic hedgehog expressing feeder cells. This result indicates that the application of the knowledge from the molecular biology is useful to develop the tissue engineering (T. Kameda, Tissue Eng, 2001). We also analyzed the nature of epidermal stem cells in hair bulge or basal layer of epidermis. Elimination of the label-retaining cells in the bulge by photo-induced cell killing method, which we developed, caused a defect on hair renewal (T. Kameda, Genes Cells, 2002). This result support the idea that the label-retaining cells in the bulge is the hair stem cell. And, we analyzed the distribution pattern of the epidermal stem cells in basal layer of mouse ear epidermis by an in vitro and in vivo analysis using retrovirus vector, and proposed new hypothesis for the epidermal renewal unit (T. Kameda, Exp Cell Res, 2003).
