Research Abstract	【複素顕微鏡】昨年に引き続き位相板の改良を行い、帯電問題の解決を図った。特にカーボン膜作成時のクリーン化法を開発した。位相差法、微分干渉法を用いて引き続き応用研究を行い、特に細胞・組織の電顕像において良好の結果を得た。【チャネル蛋白質】血管内皮細胞において、TRPC5チャネルが、一酸化窒素産(NO)産生酵素eNOSを活性化するCa^<2+>流入を担うことが示された。TRPC5チャネルとeNOSによる、Ca^<2+>とNOを介したpositive feedback loopは、内皮細胞の分化成熟にともなって形成されることも明らかとなった。【電位センサー蛋白質】電気的情報をイオンの出入りなしに化学的情報に変換する電子センサー蛋白質を見出した。原索動物ゲノムのアノテーション作業により、非チャネル型新規電位センサー蛋白を同定し、ポアドメインの代わりに酵素ドメインを有し、リン脂質の酵素活性が膜電位により変化することを明らかにした。【化学センサー蛋白質】好熱性一酸化炭素酸化細菌由来の新規なCOセンサータンパク質を新たに見い出した。大腸菌における大量発現系を構築し、精製したCOセンサータンパク質のセンサー活性中心の構造解明を行った。酸素センサータンパク質HemATにおいて、酸素の選択的センシングに関与しているアミノ酸残基を特定した。【蛋白質機能素子作製】チャンネルシリコン基板上に集積したバイオセンサー素子の作製を目標とし、素子構造を設計と要素技術開発を進めた。放射光によるシリコン表面のピンポイント微細加工、有機単分子膜をアンカーとした脂質二重膜の形成、COOH化を利用したパターと配列化技術を開発した。
