Research Abstract |
Stem cells have a remarkable potential for developing into many different cell types in the body. When a stem cell divides, each new cell has the ability to either remain a stem cell (self-renewal) or become another type of cell with a more specialized function (specific cell lineage commitment). Stem cells derived from postnatal animals (especially humans) are called adult stem cells and are generally thought to be multipotent stem cells. Among adult stem cells, mesenchymal stem cells (MSCs) are also multipotent cells that can be isolated from adult bone marrow and can be induced in vitro and in vivo to differentiate into a variety of mesenchymal tissues, including bone and cartilage. Here, we report that we have succeeded to proliferate the number of MSCs from adult human as well as rat and also induced theses MSCs into osteo/chondrogenic lineage. Importantly, the osteogenic cells in the pore regions of various porous ceramics can be used to repair osseous defects and we have used the cells for the clinical applications. Interestingly, the MSCs can also differentiate into hepatocyte and neuron. When MSCs were cultured in the presence of hepatocyte growth factor, the cells showed albumin and c-Kit expression. When the MSCs were cultured in the presence of mercapto-ethanol, the cells' morphorogy resembled neuron and showed Nestin expression. Therefore, MSCs derived from bone marrow can be used to regenerate hepatic and neural function. These results indicate the usefulness of MSCs for tissue engineering and, thus, the cells hold promise for utilization in regenerative medicine.
