Reverse SAGEを用いた新規胃癌特異的遺伝子の同定とその機能解析

| Research Abstract |
胃癌特異的に発現している遺伝子を同定することを目的し、早期胃癌1例、進行胃癌3例、胃癌のリンパ節転移1例をSAGE法を用いて遺伝子発現を網羅的に解析した。SAGEライブラリー合計約1万クローンを解析し、約14万tagが得られた。これらのデータは公的な遺伝子発現データベースであるGEOに登録している(GSE545,SAGE Hiroshima gastric cancer tissue)。これらのSAGEライブラリーのデータより、われわれはCDH17、APOE等の浸潤、転移に関わる多数のマーカーを同定した。さらに、GEOに登録されている種々の全身正常組織のSAGEライブラリーとの比較により、われわれは胃癌にほぼ特異的に発現している遺伝子としてREGIVを同定し、報告した(Cancer Res,2004)。その他では、スキルス胃癌特異的に発現している遺伝子に対してSpecifically Expressed in Scattered-type gastric carcinoma (SES)と仮名をつけ、reverse SAGE法にてクローニングを行っている。SES1については、pyruvate dehydrogenase kinase, isoenzyme 4として最近報告された。本遺伝子については胃癌特異性は報告されておらず、現在胃癌特異性をリアルタイムRT-PCRにて検討中である。SES3についてはcytokine receptor-like factor 2として同定され、全身臓器にて発現を確認したところ、多くの正常臓器で発現しており、明らかな胃癌特異性については確認できなかった。SES2についてはreverse SAGE法によるクローニングを現在行っている。いくつかの遺伝子断片を得ているが、全長のクローニングにはいたっておらず、現在も全長をクローニング中である。
