Drug development by investigation of a-synucelin aggregation mechanism through FABP

Outline of Annual Research Achievements

私達は中脳ドパミン神経に高発現する脂肪酸結合蛋白質3(FABP3)がαシヌクレインと複合体を形成し,オリゴマー形成を促進することを見出した。さらに、FABP3リガンドのなかからオリゴマー形成を抑制する低分子化合物を探索し、FABP3選択的リガンドの構造最適化を行った。FABP3の3次元構造解析から選択性・結合性の高い化合物MF1選択し、LC-MS/MSで良好な脳移行性を確認した。2018年度はαシヌクレインとFABP3の過剰発現細胞(Neuro2A細胞)でMF1のαシヌクレイン凝集抑制作用を確認した。特に、Neuro2A細胞ではアラキドン酸誘導のαシヌクレイン凝集をMF1処置は完全に抑制した。次にMPTP誘発のパーキンソン病モデルマウスを用いて、MF1のin vivoでの薬効を確認した。MF1(0.3, 1.0mg/kg)の慢性経口投与はMPTP誘発の運動機能障害を改善し、黒質におけるαシヌクレインのオリゴマー形成を抑制した。その結果、黒質におけるドパミン神経細胞の脱落を阻害した。さらに、MPTP誘発パーキンソン病マウスの認知機能障害も改善した。一方、L-DOPA慢性投与ではドパミン神経脱落と認知機能障害改善作用は見られなかった。以上の結果から、アラキドン酸がαシヌクレイン凝集に関わることを細胞系で確認した。さらにFABP3選択的リガンドMF1がパーキンソン病モデルマウスでαシヌクレインのオリゴマー形成を抑制し、運動機能のみならず認知機能も改善することを証明した。
