Outline of Annual Research Achievements

最近,骨格筋から分泌される内分泌因子:マイオカインが複数同定されており,自己作用だけでなく,脂肪細胞や骨細胞とクロストーク(相互作用)することが報告されている。しかしながら,運動や食品摂取によって誘発されるマイオカインが動脈硬化改善に関与するか否かの検討は国内外を見ても皆無である。本研究は,動脈硬化に対する運動・食品摂取効果のメカニズムに関与するマイオカインを同定し,マイオカイン作用の貢献度や分子機序の解明から効率的な運動・食事プログラムの構築を目指すことを目的とする。平成30年度では,老齢マウスを用いて,運動による動脈硬化度の改善効果に関わる候補マイオカインXの分泌部位およびマイオカイン変動が動脈硬化度の低下に対して関与しているかについて検討した。38週齢の老齢促進SAMP1マウスを老齢対照群および老齢有酸素運動群の2群に分け,12週間後に脳,肺,心臓,肝臓,腎臓,腹腔マクロファージ,骨格筋(前脛骨筋)を摘出した。Xマイオカイン遺伝子発現はreal-time PCR法にて測定した結果,他の組織よりも骨格筋で顕著に高い発現が認められた(P < 0.01)。また,有酸素性運動により骨格筋のXマイオカイン遺伝子発現は有意に変化し(P < 0.05),動脈血管の内皮機能(アセチルコリンによる血管拡張応答)の変化に対応していた。さらに,38週齢の老齢促進SAMP1マウスを用いて12週間のマルチサプリメントによる栄養摂取によっても骨格筋のXマイオカインの遺伝子発現が変化することも明らかにした。これらの結果から,候補マイオカインXは骨格筋にて発現が高く,運動や栄養摂取により変動することが明らかとなった。

Reason

ヒト試験において有酸素性運動や栄養摂取により動脈硬化度の改善効果に関わる候補Xマイオカインは動物実験においても同様の結果が得られた。今後,異なる運動様式(レジスタンストレーニング)による変化および栄養摂取との併用効果についても検討する必要性がある。

Strategy for Future Research Activity

運動や栄養摂取によるマイオカイン分泌の変化は有酸素性運動による変化は異なる運動様式(レジスタンストレーニング)によっても同様の変化を示すのか,また,栄養摂取との併用効果について検討し,運動や栄養摂取による動脈硬化に及ぼす影響にマイオカインが関与するのかについて検討する。
