Outline of Annual Research Achievements

造血幹細胞の分化・増殖により生体の成熟血球が供給される。健常な造血において、骨髄造血幹細胞はストレス造血時などに備え、自己複製能を保持し静止期状態で維持されている。本研究では、幹細胞・巨核球の産生において最重要なサイトカイン、トロンボポエチン (Thpo)シグナルがどのように、ミトコンドリア・小胞体(ER)代謝を制御し、造血幹細胞の分化・維持に関わるのか、その分子メカニズムを明らかにした。
引き続き、Tumor suppressor geneであるFolliculin (Flcn) KO mouse(研究分担者;馬場理也)とミトコンドリア膜結合分子(MAM)の一つであるMitol KOマウスを用いて、造血幹細胞におけるミトコンドリア代謝・ライソソーム機能を解析した。
さらには、「造血発生過程の解析からの造血幹細胞の誘導」に挑戦し、以下の結果を得た。胎生8-9日に、背側大動脈のhemogenic endothelial cellから造血細胞が出現する。我々の研究により、マウス胎児では、造血性血管内皮細胞(EHC)から造血幹細胞の出現とほぼ同時期に、赤芽球骨髄球前駆細胞(EMP; Erythro-Myeloid Progenitor)が検出されることが明らかとなった。今回、造血幹細胞とEMP、これら2つの細胞系譜は分離され、胎児肝においては、HSCは、もっぱら自己複製をし、分化には寄与しない、一方、成熟細胞は、前駆細胞から供給されるということを、造血幹細胞特異的に発現する遺伝子Hepatic Leukemic Factor, Hlfレポーターマウスを作製して明らかにした。また、この幹細胞の自己複製は、Evi-1転写因子の発現量に比例することを明示した。
これらのデータは、「幹細胞の自己複製と分化は切り離され得る」というきわめて重要なコンセプトと考えている(Nature、2022)。
