神経因性疼痛モデルラットにおけるP2X受容体を介した痛覚情報伝達制御機構の解析

Research Abstract

今回、神経因性モデルラットの脊髄スライス標本を用いたパッチクランプ記録を行い、脊髄後角細胞のシナプス後細胞におけるATP受容体の役割を検討した。代謝安定型のATP受容体広作動域作動薬であるATP-γS(100μM)を灌流投与すると、約40%の脊髄後角深層細胞において内向き電流と興奮性シナプス後電流の発生頻度の増強効果が観察された。一方、α,β-methylene ATP(100μM)の灌流投与を行うと内向き電流は観察されず、興奮性シナプス後電流の発生頻度の増強効果のみが観察された。ATP-γSにより生じた内向き電流は、各種P2Y受容体作動薬の灌流投与によって再現されなかった。また、細胞膜G蛋白質阻害薬存在下において、ATP-γS灌流投与による内向き電流は抑制されなかった。さらに、ATP-γS灌流投与によって生じた内向き電流ならびに興奮性シナプス後電流の発生頻度の増強効果は、P2X受容体拮抗薬であるPPADS(10μM)の存在下において完全に阻害されたが、TNP-ATP(20μM)の存在下において影響されなかった。以上の結果から、神経因性モデルラットにおいて、ATP P2X受容体は脊髄後角深層細胞のシナプス前のみならず、シナプス後細胞においても発現しており、シナプス前とシナプス後細胞に発現しているP2X受容体は異なるサブタイプであることが明らかとなった。さらに、脊髄後角深層細胞のシナプス後細胞に発現するP2X受容体は、薬理学的特性から緩徐脱感作型のP2X受容体であることが示唆された。
