Outline of Final Research Achievements |
The aim of the project was to decipher mechanisms underlying metabolic changes in glomerular podocytes in health and disease. To meet this aim, firstly, in vitro cell culture systems were exploited. We chose the MDCK cell as a model system since it is an excellent model for the renal epithelial cells. Utilizing CRISPR/Cas9 genome editing technology, genes encoding mammalian Ajuba family proteins were knocked out in MDCK cells. The stable MDCK line lacking LIMD1 has been successfully established.

Academic Significance and Societal Importance of the Research Achievements |
Ajuba関連遺伝子は細胞間のイオン透過の調節に不可欠な接着結合に局在することが報告されている。接着結合は腎臓においても決定的な機能を担うことから、樹立された細胞株は腎臓による体液の恒常性の維持を研究する上で有用なツールとなることが期待される。
