Outline of Annual Research Achievements

パーキンソン病サルの特定の脳部位にウイルスベクターを注入し、治療することを計画している。そのため、ターゲットになる脳部位を同定し、そこから神経活動を記録することを行った。海外共同研究先であるスペインCINACを訪問し、以下の共同実験を行った。2頭の正常カニクイザルを用い、全身麻酔下で頭部を無痛的に固定するための手術を行った。手術の翌日、頭部を無痛的に固定し、一次運動野、補足運動野周囲の頭蓋骨を除去し、これらの大脳皮質の生理学的マッピングを行なった。マッピングに基づき、一次運動野、補足運動野の上肢領域に刺激電極を埋め込んだ。さらに、淡蒼球外節、内節、視床下核に金属記録電極を刺入し、神経活動記録を行なった。自発活動、身体各部位の刺激に対する反応などに加え、大脳皮質を電気刺激した際の応答について刺激前後時間ヒストグラムを作成して調べた。その結果、淡蒼球外節、内節は、高頻度自発発射と大脳皮質刺激に対する早い興奮―抑制―遅い興奮から成る3相性応答を示し、視床下核は、中頻度自発発射と大脳皮質刺激に対する早い興奮―遅い興奮から成る2相性応答を示した。これらの特徴的な反応によって、淡蒼球外節、内節、視床下核の上肢領域を同定することができた。
また、日本においては正常ニホンザルを用いて、視床下核をブロックすることを試みた。具体的には、上記と同様な方法を用いて神経活動を記録することにより、視床下核を同定した。次に、抑制性のDREADDであるhM4Diを搭載したAAVウイルスベクターを同定した視床下核に注入した。hM4Diの発現を待って、hM4DiのリガンドであるクロザピンN-オキシドを全身投与したところ、ヘミバリスム様の不随意運動を示した。これらのことから、サルに対してDREADD法を用いることにより、視床下核をブロックできることがわかった。

Reason

海外共同研究先でサルを用いた神経生理学的実験ができる準備を行い、実際、実験を行ったこと、日本においても正常サルではあるが、大脳基底核をDREADDによってブロックできることを示すなど、実験計画が順調に進んでいる。

Strategy for Future Research Activity

海外共同研究先でのサルの大脳基底核マッピングに関する共同実験を継続するとともに、日本においてはパーキンソン病モデルサルを用いて、視床下核ブロックをする実験を開始する。

Causes of Carryover

共同研究先での実験が中心であり、実験費用などは先方持ちであったので、次年度使用額が生じた。翌年度分と合わせて、視床下核ブロック、大脳基底核記録にむけての消耗品や研究員の雇用に使用したい。
