Outline of Final Research Achievements |
Cells generally undergo replicative senescence when cultured for long periods. Senescent cells secrete senescence-associated secretory phenotype (SASP) factors, such as inflammatory cytokines, causing deleterious effects on surrounding tissues. In this project, we aimed to collect basic knowledge that will serve as a foundation for stem cell therapy by (1) characterizing senescent dedifferentiated fat cells (DFAT cells) and (2) exploring effective methods of removing senescent cells to enhance bone regeneration. As a result, we observed the flattened shape of cells, nuclear enlargement, and expression of several senescence markers and SASP factors after repeated passages of DFAT cells. Although we could not reach the second project during the entire research period, we reached a detailed understanding of the characteristics of replicative senescent DFAT cells.

Academic Significance and Societal Importance of the Research Achievements |
均質で膨大な量の細胞の確保は、今なお再生医療の発展における足かせとなっている。組織採取が容易な脂肪に起源を持つ脱分化脂肪細胞(DFAT細胞)は、骨髄由来間葉系幹細胞(BMSC)同様に増殖能に優れ、新たな細胞ソースとして期待されている。本研究では、細胞老化後のDFAT細胞の詳細な特性の解析に取り組んだ。その結果、細胞形態の扁平化や、核の肥大化、複数の老化マーカー・種々のSASP因子の発現増強が確認できた。
