Outline of Final Research Achievements

Through our series of study, it was confirmed that organoids derived from functional neuroendocrine tumors can be established from multiple different cases using established methods. Additionally, the cultivation progress and proliferation efficiency after establishment showed varying patterns depending on individual cases, suggesting a reflection of the gene expression patterns in the original tissues. Moreover, based on the gene expression patterns obtained from analyses using organoids derived from functional neuroendocrine tumors, candidate factors that regulate the expression of specific secreted proteins were identified. Comparative verification was also conducted regarding the appropriate models for transplanting these organoids, and conditions under which engraftment can be achieved with a certain efficiency were confirmed.

Academic Significance and Societal Importance of the Research Achievements

特定の分泌蛋白について恒常的な発現制御機能を欠くとした機能性神経内分泌腫瘍について、同腫瘍由来のオルガノイドを樹立・解析するための手法を確立した。また複数症例の機能性神経内分泌腫瘍由来オルガノイドを比較検討することにより、機能性分泌蛋白に関する異なる制御機構を反映した遺伝子発現パターンに関する網羅的データの取得に成功し、これに基づき発現制御因子群を抽出する基盤を確立した。
