Outline of Final Research Achievements

To reveal functional cooperation, we generated alpha-synuclein and tau double-knockout mice and characterized the functional cross talk between these proteins during brain development. As result, we find that deletion of alpha-synuclein and tau break the balance between the proliferative and neurogenic divisions of neuronal progenitor cells, resulting in an overproduction of early born neurons. On the other hand, precocious neurogenesis causes reduction of the number of neural progenitor cells in the middle stage of corticogenesis to diminish subsequent gliogenesis, and lead to reduction of brain size after born.

私たちはアルファシヌクレインとタウのDKOマウスの解析を通して生理的な機能を明らかにしたが、DKOマウスに見られる神経形成の異常はこれまで報告されているものと全く
異なったメカニズムで小脳症が発症している。したがってアルファシヌクレインとタウの関連研究は神経発生と神経変性の新たなフロンティアの開拓につながることが期待される。このことはアルファシヌクレインとタウの生理的な機能に迫る大きな手がかりを初めて提示するものである。
