Outline of Final Research Achievements

In this study, we first searched for genes that could be super-enhancers and selected candidate genes. We then analyzed the expression of this gene family in dental pulp-derived mesenchymal cells, odontogenic epithelial cells, and various organs, and found that it was specifically expressed in teeth and the brain. We also analyzed the expression of the gene during tooth embryo development by in situ hybridization. The results suggest that the gene are expressed only in a limited stages and may play a role in the development of tooth embryos.

Academic Significance and Societal Importance of the Research Achievements

組織再生は周囲の多くの細胞の相互作用によって営まれるが、このプロセスにおける間葉系幹細胞と線維芽細胞の相互作用は明らかとなっていない。本研究において、創傷治癒ではたらく線維芽細胞と間葉系幹細胞(MSC)の関係を明らかにし、特に細胞分化の鍵を握るスーパーエンハンサーを中心とした新規組織再生法の開発を行うことで、歯髄中にも豊富に存在する線維芽細胞から直接的に象牙芽細胞への分化を促すダイレク トリプログラミング法を開発できれば、象牙芽細胞やエナメル芽細胞への分化を飛躍的に誘導する新規の細胞誘導法の開発へも発展できると考える。
