タンパク質の精密ナノ凝集制御界面での細胞の接着と機能

Outline of Annual Research Achievements

吸着タンパク質を制御するため両親媒性ブロックコポリマーを用いた.両親媒性ブロックコポリマーは親水性部位と疎水性部位の自己組織化によりナノスケールの相分離構造表面をつくる.タンパク質吸着を抑制することで知られるpoly (2-methacryloyloxyethyl phosphorylcholine) (PMPC)を親水性部位として含んだ両親媒性ブロックコポリマーを用い,疎水性ドメインがドット状となるナノ相分離構造表面上では,疎水性ドメインに選択的にタンパク質が吸着することが報告されている.そこで本研究ではPMPCと疎水性のpoly(3-(methacryloyloxy)propyltris(trimethylsilyloxy)silane) (PMPTSSi)からなる両親媒性ブロックコポリマーを用い,疎水性部位がマトリックスを形成している表面とドット状ドメインを形成している2種類のタンパク質の吸着分布状態が異なる表面を作製した.相分離構造の観察はTEMを用いて行った.また構造観察には、各ポリマーをコーティングした基板をPBSに一晩浸漬し、その構造をAFMによりタッピングモードで形状観察を行った.まず大気中での相分離構造を観察し,その後フィブロネクチン溶液(10 μg/mL)に浸漬させた表面も同様に大気中で観察した.Siカンチレバー(曲率半径: 7 nm, ばね定数: 42 N/m)を用いて測定を行った.さらに表面に吸着したフィブロネクチンの量を測定した後、serum free mediaでマウス繊維芽細胞を播種し接着性の評価を行った.疎水性表面に吸着したタンパク質の分布を、AFMを用いて解析することができた。また、タンパク質吸着量が同じであっても、その分布状態の違いにより細胞接着挙動に違いが認められた。

Strategy for Future Research Activity

タンパク質の吸着分布状態が異なることがわかったため、次年度以降は、その吸着したタンパク質の構造変化についてを解析し、より細胞接着メカニズムの解明へとつなげる。
