二弁性大動脈弁における神経堤細胞の役割とその分子基盤の解析

Outline of Annual Research Achievements |
心臓の半月弁、すなわち大動脈弁と肺動脈弁の形成は、心内皮由来間葉細胞、神経堤細胞由来細胞、二次心臓領域由来細胞が関与しているといわれている。弁周囲は心筋層と血管平滑筋層の両方が周囲環境としてある。最終年度では、ウズラ-ニワトリキメラを作成し、大動脈弁、肺動脈弁だけでなく大動脈弓の途中まで神経堤細胞が分布していることを見出した。神経堤マーカーを示す遺伝子改変マウス(Wnt1Cre-マウス)とR26REYFP-マウスとを交配し、流出路で蛍光発現する細胞1個ずつの性質を見極めたところcKitが神経堤細胞と関連していることを見出し、現在も検討中である。また流出路の冠血管入口部にSemapholin3E-PlexinD1受容体のシグナルが働いていることを見出し、発生途中の流出路大血管周囲に形成される脈管とリンパ管の異常がSemapholin3E ノックアウトマウスに見られた。本ノックアウトマウスでは心血管の異常も認めた。
