Research Abstract

有効かつ安全ながんの遺伝子・核酸医薬による治療法の実現に向けて、アテロコラーゲン・デリバリーシステムによる生体内遺伝子発現制御法を開発した。この方法はバイオマテリアルの一種であるアテロコラーゲンをキャリアとして特に核酸医薬を生体内にデリバリーし、ヌクレアーゼによる生体内分解から核酸を保護することで、遺伝子ベクターの欠点であった生体内での非制御性を改善し、ベクターの能力を最大限に発揮させうる画期的な方法である。本年度はこのデリバリー技術を用いて,アテロコラーゲンとmicroRNAの複合体は、siRNAの場合と同様に、細胞に取り込まれやすいナノサイズの粒子径であり、生体内でのmicroRNAの安定性に大きく寄与し、ヒト前立腺がんで顕著に低下している事が判明したmicroRNA16の補充療法に有効である事を示した。さらに、このデリバリーの担がんマウスにおける腫瘍内での導入と消失を検討した結果、全身性の投与後、2日後に腫瘍内にmicroRNAが確認され、その濃度は3、4日と徐々に上昇し、最大値に達した後、2から3日間はその値を持続した後、ゆっくり消失し、10日後には確認不可能となった。他の正常臓器では、これとは対照的な動態となり、例えば肝臓では投与直後に正常値の2-3倍となったが、その6時間後には元の値にまで戻っていた。この知見は、今後、microRNAの補充療法におけるmiroRNAの体内動態を考える上で重要な参考になると考えられた。以上の解析から、アテロコラーゲン包埋方法の応用は、microRNAにまでその効果が及ぶ事が示唆され、今後のnon-coding RNAによる制がん研究に本法は有用であると考えられた。
