Outline of Research at the Start

造血幹細胞は、骨髄中の様々な周囲細胞(ニッチ)に分化と増殖が制御されている。 動脈硬化の発生や増悪に重要な単球とその前駆細胞も骨髄中でニッチに囲まれ、制御されて いることが研究代表者の研究で明らかになった。本研究の目的は、単球の分化とその制御に 関わる骨髄ニッチに着目し、動脈硬化形成時に起こる変化を解明し、骨髄をターゲットとし た新規動脈硬化予防法を開発することである。

Outline of Final Research Achievements

We tried to clarify atherosclerosis specific monocyte population in blood and bone marrow by single cell RNA sequencing. Although we fined an increase of monocyte population in bone marrow, we could not find atherosclerosis specific cell populations in bone marrow or bone marrow niche.
Single-cell RNA sequencing revealed more monocytes, mast cells and inflammatory macrophages accumulated in coronary plaques involved in acute coronary syndrome in comparison to culprit plaques for chronic coronary syndrome.

Academic Significance and Societal Importance of the Research Achievements

動脈硬化における骨髄での単球増加を示すことができた。特異的な細胞集団の発見にまではいたらなかったが、骨髄の単球造血が動脈硬化に与える影響の一端を示すことができたと考える。
