Outline of Research at the Start |
先行研究において2次元培養でのiPS細胞とヒト由来の歯肉線維芽細胞と共培養した心筋組織の作成に成功した。この結果をもとに、生体へ臓器を移植するためのステップとして、より巨大な組織の作成が必要と考えられた。本研究では異種動物由来材料の非存在下でiPS細胞を3次元的環境下にて分化誘導し、立体的な心筋組織を作成する。本研究の成果は、虚血性心疾患患者に対するドナー心臓不足の解消につながることが期待される。

Outline of Final Research Achievements |
In this study, for the purpose of creating mature myocardial tissue, human-derived iPS cells and human gingival fibroblasts were co-cultured, and stretch stimulation was applied by a cultured cell extension system in the process of inducing differentiation into cardiomyocytes. The prepared myocardial tissue was subjected to qPCR for evaluation at the gene level and immunostaining for evaluation at the protein level. Furthermore, for morphological comparison of the fine structure of the tissue, observation was performed using an electron microscope, and a moving image of the tissue was taken to analyze the contractility. As a result, it was suggested that stretch stimulation may affect the maturation of iPS cell differentiation into cardiomyocytes.

Academic Significance and Societal Importance of the Research Achievements |
心臓移植は、薬物療法と機械的補助療法だけでは治療が困難であった末期心不全の唯一の治療法である。 iPS細胞で心臓を作ることが可能になれば、ドナー不足の問題は解決される。これまでiPS細胞の心筋組織への分化を誘導するための様々な方法が考案されており、本研究では、ストレッチ刺激を適用することにより、iPS細胞の心筋組織への分化の成熟に影響を及ぼす可能性が示唆され、将来的な臨床応用に向けて前進していると考えられる。
