研究概要

電位依存性カルシウムチャネルβサブユニットの交感神経制御機構における重要性を, 遺伝子改変動物を用いて解析を行った
交感神経による心筋収縮制御機構はカルシウムチャネルを介して行われている. 心筋において最も発現している電位依存性カルシウムチャネルβサブユニットはβ2である. この遺伝子の欠損マウスモデル(β2サブユニット欠損マウス)を作製し, 生きた個体を用いて, 交感神経への影響を調べた. β2遺伝子欠損マウスにおいては交感神経刺激による心筋収縮の増強作用が低下していることを確認した. さらに電流量の増加反応が認められないことを確認した. 電位依存性Caチャネルβ2サブユニットのリン酸化部位に変異を導入し, 変異遺伝子を過剰に発現するマウスモデルを作製した.(Murakami et al., 現在論文投稿中)
