Outline of Final Research Achievements |
As PRL1 was not an efficient therapeutic target localizing at cell membrane of castration-resistant prostate cancer (CRPC), we alternatively focused on KIFC1, a member of kinesin-14 family that involved in cell division and intracellular transport. KIFC1 was involved in acquired resistance to taxane anticancer agents in CRPC cells. Antitumor activity of docetaxel to taxane-resistant CRPC cells was recovered only when concomitant administration of KIFC1 inhibitor drug, CW069. No cytotoxic actions of CW069 to normal prostate cells were identified. Therefore, KIFC1 inhibitor drug, CW069 might be promising therapeutic drug for CRPC.

Academic Significance and Societal Importance of the Research Achievements |
去勢抵抗性前立腺癌(CRPC)の治療薬の開発は近年目覚ましいものの、いまだ治癒は難しく、死に至る病である。CRPCが予後不良な疾患である要因の一つに、効果を示した薬剤がいずれは効かなくなる、つまり耐性を獲得することが挙げられる。本研究により、CRPC細胞がタキサン系抗がん剤への耐性を獲得する過程にモーター蛋白であるKIFC1が関与し、さらにKIFC1の阻害薬の投与でタキサン系抗がん剤への耐性が緩和され、抗腫瘍効果が改善することが示された。一度耐性を示した治療薬にもう一度感受性を取り戻させることができるという、既存の薬剤機序とは異なったアプローチであり、有望な治療法となる可能性が示唆された。
